If I have a direct contract with a homeowner can I still lien their property even if I never sent an NTO?

A Florida Notice to Owner is required from any party wishing to file a mechanics lien that doesn't have a direct contractual relationship with the property owner. This is one of the few exceptions to the NTO requirements. The next step would be to file a mechanics lien claim in the county clerk's office where the property is located.
However, one useful tool before filing a mechanics lien is to send a Notice of Intent to Lien (NOI). This isn't a required form in FL, but merely a letter stating that if payment or a payment plan isn't established within a certain amount of days, that a lien will be filed on their property. This is typically enough to loosen the grip on your cash.
