As a truck driver I drove under contract and the owner refuses to pay. Can I put a lien on his vehicle?

No, you cannot file a a lien. Chapter 53 liens only apply to improvements to real property, not services (driving a truck). Sorry. Your only option is to sue him for non-payment.
