Can I file a lein on a house I co-signed on with an exboyfriend whom in turn is suppose to help me on building a deck for my house. My signature on his house in exchange for him helping me with my deck. He is a General Contractor.

Generally, mechanics liens will only be available to those who have performed work, themselves, which has permanently improved the property to be liened. So, contractual agreements or financial help won't give rise to mechanics lien rights - though performing labor or furnishing materials to the property will.
With that being said, if there's a contract or agreement in place and if the other side of that agreement isn't holding up their end of the bargain, then it might be possible to pursue other claims against them - or to at least threaten to make a claim in order to try and have them act in accordance with the contract.
