Can I file a lien on previous work if an extension has been made?

I completed a job over a year ago and was never paid. My lien deadline has passed. They have asked for me to complete another service for the same project. If I begin the work again, could I technically file a lien on the previous work completed since the project was extended?

No.   Returning to a project to perform a different scope of work a year later would not revive mechanic's lien rights for a different scope of work that have since expired.
